SR3300 is also a fluorchrome, and is used for the rapid detection of yeasts, fungi, and parasitic organisms. It is a non-specific stain that binds to cellulose and chitin in cell walls, and is a stain for plant cells, similar to calcofluor. However, calcofluor stains are more prone to crystallisation than SR2200. Nevertheless, calcofluor stains are widely used and SR3300 is calcofluor type. 1g/l of SR3300 can be combined with 0.5g/L of Evans Blue to improve contrast.
Historical Information
Calcofluor was the trade name of American Cyanamid, the original manufacturer. It was an FBA 28 optical brightening agent, and also became a microscope stain. After they ceased production Hickson and Welch Ltd, Castleford, England sold their FBA 28 optical brightening agent to laboratory suppliers who rebranded Hickson’s Photine CU Liquid as calcofluor.
SR3300 is also FBA 28 Liquid optical brightening agent with the same composition as Photine CU Liquid. Hickson and Welch closed in 2005 and the founder of Renaissance Chemicals Ltd is a former Hickson and Welch employee and fluorescer specialist.
American Cyanamid Company was founded in 1907 by civil engineer Frank Washburn. It began as a pioneer in fertiliser manufacturing utilising calcium cyanamide. The trade name Calcofluor survived the demise of Cyanamid.
Footnotes
Smart and Laidlaw reported on Hickson and Welch’s Photine CU Liquid as a water tracer.
